AB OBJECTIVE: Clarification of the roles of Human T-cell leukemia virus
type-1 (HTLV-1) in HTLV-1-associated myelopathy (HAM)/tropical spastic
paraparesis (TSP) with rat model of HAM/TSP. METHODS: HTLV-1-producing T
cells were injected intravenously into WKA and F344 rats. Detection of
HTLV-1 provirus in peripheral blood mononuclear cells and organs of the
rats was performed by PCR. Titers of anti-HTLV-1 gag and envelope
antibodies were analyzed by particle agglutination kit and ELISA,
respectively. RESULTS: Six out of the eight WKA rats injected with
HTLV-1-producing T cells showed HAM/TSP-like paraparesis, while no
symptoms were observed in seven F344 rats injected with HTLV-1-producing
T cells or eight untreated WKA rats. The antibody response against
envelope protein was lower in WKA rats than in F344 rats. White matter
degeneration with infiltration of macrophages was found in the spinal
cord and peripheral nerves. DISCUSSION AND CONCLUSIONS: This study
showed the importance of HTLV-1 infection and the genetic background of
the host in the neuropathogenesis of the HAM/TSP-like rats.
